PCR (Polymerase Chain Reaction) needs primers to start DNA synthesis.
These primers are short single-stranded DNA sequences (not polypeptides).

direction, so the primer must bind to the 3 '-end of the template strand to allow extension. Therefore, primers are designed to be complementary to the 3 '-ends of the DNA region to be amplified.
