Calcium treatment(enhances cell permeability), microinjection (directly inserts DNA), and biolistics (shoots DNA-coated particles into cells) are techniques used to modify cells in such a way that DNA can be introduced into cells. PCR, Agarose gel electrophoresis and spooling are not for introducing rDNA into cells. PCR is used to amplify DNA, agarose gel electrophoresis is used to isolate and separate DNA and spooling is used to separate out DNA.
