pBR322 is a commonly used cloning vector. Here, gene coding for beta-galactosidase is inserted in the plasmid pBR322 at PstI restriction enzyme site which is responsible for providing resistance toward antibiotic ampicillin. This causes insertional inactivation of the antibiotic resistance gene thus E.coli strain will not provide resistance towards the antibiotic ampicillin
